Comparison of Selected Commercial Sugarcane Varieties for Use in a Stable Transformation Programme
نویسنده
چکیده
Ten commercial sugarcane varieties, N12, N14, N19, N24, N25, N26, N27, 75E247, 84E1334 and NCo376 were tested for suitability for transformation in terms of their embryogenic callus production and transient expression levels. Based on an increase in callus mass over time, varieties N12, N19, N27, NCo376 and 84E1334 outperformed other varieties on Murashige and Skoog medium containing 2,4-dichlorophenoxyacetic acid (3 mg/L). Using microprojectile bombardment and a plasmid which carries the maize anthocyanin gene, transient expression levels of varieties were determined by counting the number of red foci per cm2 callus material 48 hours after delivery. Varieties NCo376, N12, N27 and N19 showed high transient expression levels of between 1 000 and 1 450 red foci per cm2 callus material. Regeneration regimes for varieties differed and these are discussed. Three varieties, NCo376, N12 and N19, were selected for further stable transformation studies on the basis of promising performance.

EVIDENCE FOR DIFFERENT NITROGEN USE EFFlClENClES OF SELECTED SUGARCANE VARIETIES
A rapid screening (hydroponic) and field testing programme There is increasing interest in improving the nitrogen (N) use efficiency of cultivated crops, due to the high cost of synthetic N fertiliser. Since existing differences in N use efficiency between sugarcane varieties could be exploited to improve N fertilisation, a hydroponic pot experiment was conducted to test the performance of seve...
متن کامل2G ethanol from the whole sugarcane lignocellulosic biomass
BACKGROUND In the sugarcane industry, large amounts of lignocellulosic residues are generated, which includes bagasse, straw, and tops. The use of the whole sugarcane lignocellulosic biomass for the production of second-generation (2G) ethanol can be a potential alternative to contribute to the economic viability of this process. Here, we conducted a systematic comparative study of the use of t...
متن کاملSugarcane (NCo310) Transient Transformation Using uidA Reporter Gene
Background: Sugarcane is a monocotyledonous crop that is cultivated in the tropical and subtropical regions of the world. One of the most important criteria, influencing the efficiency of the sugarcane transformation is known to be related to physical and biological factors during the transformation procedure. Objectives: The objective of this research was to study the response of callus induc...
متن کاملThe Identification of Potential Genetic Markers in Sugarcane Varieties for the Prediction of Disease and Pest Resistance Ratings
The use of genetic markers that are linked to specific traits in sugarcane has the potential to increase the efficiency of the selection of improved varieties. Conventionally, markers are identified by analysing the segregation of potential markers and traits in the progeny of single crosses. However, this approach is not practical for sugarcane breeding programmes where replicated, well charac...
متن کاملUse of differential levels of mean observed heterozygosity in microsatellite loci of commercial varieties of sugarcane (Saccharum spp).
In this study, we measured the genetic diversity within and among a set of 9 commercial sugarcane varieties used for alcohol and sugar production using 17 microsatellite DNA markers. The UGSM148 and UGSM59 primers were monomorphic for all 74 sugarcane samples. The estimated proportion of simple sequence repeated (SSR) polymorphic loci was 88.23%; 17 alleles were detected. The mean gene diversit...
